Satoh was born on 21 March in Iwatsuki-ku, Saitama. Satoh briefly worked as a child actor for three or four years, appearing in commercials and magazine photo shoots. After he entered higher grades in elementary school, he decided not to continue as a child actor due to a "shy personality". Satoh was scouted by an agent from Amuse, Inc. In , he guest-starred in Shinigami no Ballad Kentaroh Ishihara and gained popularity in the seventeenth installment of the Kamen Rider series as Ryotaro Nogami. Satoh attributes the popularity of Den-O to its comedic timing.

Takeru Satoh has portrayed the character of Kenshin Himura since , and has been doing all his own stunts since he first took on the role. And to mark the franchise coming to a close with the fourth and fifth films, The Final and The Beginning, the actor has shared a closer look at how he prepares for the part. Keishi Otomo's film adaptations of Nobuhiro Watsuki's classic manga series features incredibly well choreographed fight scenes between Satoh's Kenshin and his co-stars. Kenshin is an expert in "Hiten Mitsurugi-ryu," a sword technique that allows the fighter to attack multiple assailants at once. And, keeping in line with the source material, Satoh is regularly put up against multiple actors that he has to fend off through a series of intricately detailed moves in all five films. Embodying his samurai character, Satoh reportedly spends weeks perfecting the sword fights with choreographers, going through the moves multiple times both in and out of costume in a studio before heading out on set.

He was diagnosed with pneumothorax during filming, citing chest pains, but has since recovered. After the end of his tenure as Ryotaro, Satoh continued to star in multiple dramas and films, quickly expanding his popularity within the Japanese entertainment industry. As Toei could no longer afford to hire him for reprisals, Ryotaro would later be re-written to accommodate for Satoh's absence from the role. He would be subsequently replaced by Takuya Mizoguchi , who would portray the younger " Kotaro " in the character's future appearances going forward.
