When under the servitude of Takigakure as an elite ninja, Kakuzu took on a mission to assassinate Hashirama Senju , the First Hokage. This mission, however, ended in failure against the God of Shinobi. Despite the insurmountable danger faced on the mission, what awaited his return to his village was the stigma and harsh punishment associated with the failure of his objective. In response to this treatment, a wrong levied against a shinobi who had put life and limb on the line for his village, Kakuzu came to hold a violent hatred for Takigakure as he escaped from imprisonment. In his flight from the village, Kakuzu killed the village elders, took their hearts, and fled with knowledge of the village's most prized forbidden techniques. Some time after his defection, he became affiliated with, and joined, the criminal organisation, Akatsuki. During his early years in Akatsuki, he had at least four partners that he eventually ended up killing due to his short temper, and would come to use their hearts for his Earth Grudge Fear.

The group possessed many fearsome fighters, with some even obtaining the secrets to immortality. However, out of the four members that had gained permanence, Kakuzu was the only one that was organically immortal. The secret to his eternal life lies in the forceful extraction of his enemies' organs, and while it may sound quite simple, the process certainly isn't. Kakuzu was the oldest member of the Akatsuki, dating back to the times of the First Hokage. Since then, he had prolonged his life through the hearts of his defeated opponents, and while his eternal life still had several limitations, he could increase his lifespan as long as he had the bodies of his victims. However, Kakuzu's immortality had several intricacies to it, adding up to his organic yet gruesome permanence. Kakuzu's immortality, along with his villainous origins, can be traced back to his time in the Hidden Waterfall Village. Once an elite ninja of his home village, Kakuzu was deemed a failure when he failed to assassinate the First Hokage, Hashirama Senju, prompting ridicule and disappointment from his own people. Despite his loyalty to the village, the elders punished Kakuzu for failing the mission by imprisoning him, giving way to disdain and contempt to creep into his heart. Angered by the brutal punishment and harsh treatment, Kakuzu betrayed his nation and fled the Hidden Waterfall Village, though not without slaughtering the village elders and stealing their forbidden techniques. One of these jutsu was the Earth Grudge Fear technique , a prized jutsu of the Hidden Waterfall, capable of granting the wielder near-permanence. Through the Earth Grudge Fear technique, Kakuzu could transform his body into hundreds of thick threads, with his limbs almost sewed together with these needle-like appendages. The jutsu was once a secret technique belonging to the Hidden Waterfall Village, though it was later stolen by Kakuzu once he defected from the village. These threads are capable of detaching limbs from the body, which can then be sewed back by the thick tendrils. Kakuzu could implant up to five beating hearts into his body at the same time, and for him to be slain, all five of his hearts had to be destroyed.

Kakuzu this would normally be fatal, Kakuzu had five hearts now four in his kakuzu, and thus recovered fairly quickly, sending a counter-attack to Kakashi. He then caught the rest of Team 10 with his threads, hoping to replenish his stock, kakuzu. He and Hidan slaughtered most of the other monks save one who informed the Konoha ninja of the two Akatsuki.

Kakuzu was a mercenary in the Akatsuki and among the organization's deadliest members. Able to command all releases and with five hearts in his chest, he was known as the "immortal ninja. Though Kakuzu has demonstrated proficiency in defeating dozens of enemy shinobi at once, he is not invincible. Through matching him against the series' many diverse characters, we can better appreciate the extent of his strength and the limitations that come with it. While Deidara's explosions were formidable in themselves, his deadliest and most viable ability was through implanting his targets with microscopic explosives and blowing them up from the inside out. However, these bombs can be disabled through a formidable electric charge demonstrated through his battle against Sasuke Uchiha. Since Kakuzu has a lightning release, he would diffuse himself before knocking his opponent from his mount with wind and finishing him off in melee combat.
